      3種食源性細(xì)菌免疫磁珠聯(lián)合ATP發(fā)光檢測(cè)技術(shù)研究

      沙門(mén)氏菌、單核細(xì)胞增生李斯特菌和金黃色葡萄球菌是食品中常見(jiàn)的致病菌,在國(guó)內(nèi)外常引起食源性疾病。為解決食源性疾病中的微生物檢測(cè)問(wèn)題,本研究將免疫磁珠技術(shù)和ATP發(fā)光技術(shù)聯(lián)合用于食品微生物檢測(cè),選擇沙門(mén)氏菌、單核細(xì)胞增生李斯特菌病和金黃色葡萄球菌為檢測(cè)對(duì)象,建立快速檢測(cè)這3種常見(jiàn)食源性病原菌的富集及檢測(cè)新方法。該方法在顯著縮短預(yù)增菌時(shí)間的條件下,通過(guò)免疫磁珠的富集作用獲得與常規(guī)增菌時(shí)間同樣的效果,樣品中的微生物濃度增加了10倍,大大提高了樣本檢出率,縮短了檢測(cè)周期。結(jié)果表明,本研究建立的免疫磁珠富集后聯(lián)合ATP發(fā)光技術(shù)具有快速、敏感、特異和低廉的優(yōu)點(diǎn),可用于檢測(cè)食品中沙門(mén)氏菌、單核細(xì)胞增生李斯特菌以及金黃色葡萄球菌,具有較好的推廣應(yīng)用前景。

      Chemiluminescence for Detection of three Kinds of FoodborneBacteria

      At present,SalmonellaListeria monocytogenes and Staphylococcus aureus arecommon pathogenic bacteria in food,usually causing foodborne diseases. In order to solvethe problems of microbial detection,the immunomagnetic beads technology and ATPluminescence technology were combined,and a new method was established for rapid detectionof the three foodborne pathogens. Results showed that,under the condition of significantlyshortening the pre-enrichment time of bacteria,the same effects as the conventional enrichment timewas obtained by the immunomagnetic beads,and the microbial concentration of the samples wasincreased by 10 times,which greatly improved the sample detection rate and shortened thedetection cycle. In conclusionthe established method in this study was rapid,sensitive,specific and low-cost in detection of Salmonella,Listeria monocytogenes and Staphylococcusaureus in foodand it would have application prospects.
