      MALDI-TOF-MS用于微生物鑒定的影響因素分析

      影響MALDI-TOF-MS微生物鑒定的因素有很多。本研究主要從微生物培養(yǎng)基、培養(yǎng)狀態(tài)、菌體前處理、上樣量等方面進(jìn)行MALDI-TOF-MS操作技術(shù)和方法的探討。研究結(jié)果顯示:進(jìn)行MALDI-TOF-MS細(xì)菌鑒定,培養(yǎng)時(shí)間在4~24 h之間為佳;盡量選擇無色、不含鹽的液體培養(yǎng)基;0.1 μL菌液是最低鑒定菌量;上樣量對(duì)結(jié)果無影響;對(duì)于菌體前處理,除按照標(biāo)準(zhǔn)化前處理程序操作外,也可將菌體加ddH2O制成混懸液進(jìn)行點(diǎn)靶鑒定,這樣更加簡便。本研究有助于減少MALDI-TOF-MS操作盲目性,提高鑒定質(zhì)量,利于操作的規(guī)范化和簡便化。


      Influence Factors Analysis onMALDI-TOF-MS in Microorganism Identification

      Various factors could influence the effectof MALDI-TOF-MS in microorganism identification. In this studythe approaches and techniques ofMALDI-TOF-MS from aspects of microbiological medium,bacteria incubation statebacteria pre-treatment and sample loadingquantity were summarized and discussed. According to the research results,the period of bacterium incubation of 4 to24 hours was the bestcolorless and salt-free medium was preferable;the lowest ascertainable bacteria solutionwas 0.1 μL;the sample loading quantity had no effect onidentification result. Besidesfor the standard pre-treatment procedures,a handier way was to add ddH2O into the bacteriato  make suspension and conduct targetidentification. This study would contribute to reduce the operational blindnessof MALDI-TOF-MS,enhance the identification quality andfacilitate the standardization and simplification of operation.
